Test Principle
Sperm Chromatin Dispersion (SCD) Method: This test utilizes the differential response between spermatozoa nuclei with fragmented DNA versus those with intact DNA. Controlled denaturation followed by nuclear protein extraction creates partially deproteinized nucleoids where DNA loops expand, forming chromatin dispersion halos. Spermatozoa with fragmented DNA either develop minimal dispersion halos or none at all.
Reference Values & Identification
Normal Reference Value: Percentage of sperm with fragmented DNA < 25%
DNA Fragmented Sperm Identification: Sperm heads produce small or no halos, with halo thickness on one side less than 1/3 of the smallest diameter of the sperm head
